Excited by the idea of becoming a scientist since I was a small child, my interests in pursuing graduate studies intensified during my undergraduate education and my research experience. Rather than discouraging me with the tremendous amount of work and demands for creativity, my college years motivated me to earn a Ph.D. and to continue pursuing a career as a research scientist. Although my exact research goals have yet to be refined, my interests include studying the genetic basis of disease and the genetic mechanisms that govern various cellular processes, such as the cell cycle, apoptosis, and tumor formation. Although I entered college without a clear research direction, the Biotechnology Program at Rochester Institute of Technology helped develop my interest in the field of Genetics. I found much of my laboratory training at RIT to be extremely rewarding. For example, one experience that early on convinced me

to seriously consider genetics was a project involving the construction and characterization of a recombinant plasmid. Since I worked in dependently for the most part, the project played a key role in increasing my confidence and helping me to understand various concepts pertaining to genetic analysis. It was this experience that motivated me to seek a position as a teacher抯 assistant for a course in Microbial and Viral Genetics. As a TA, I learned how demanding the teaching situation is and how much teaching a course can contribute to my own learning. By this time, after discovering my passion for research and the joy of teaching, I began to seriously contemplate graduate schools. To excel in graduate school, I decided I would not limit my experience to my formal education at RIT. Instead, I wanted the opportunity to explore my career options to make a well-informed decision as to the area of my future research. With these goals in mind, I chose to take advantage of RIT 抯 Cooperative Education Program, which meant earning my Bachelor抯 degree in five years instead of four. In December of 1997,Astra Arcus US A Pharmaceuticals hired me as a research assistant in the Electrophysiology department. At Astra, I learned research techniques not taught by RIT. For the past year, I have been working with a subset of neuronal glutamate receptors via voltage clamping on Xenopus oocytes. The research of our lab is geared toward the study of central nervous system diseases. Working with Astra and earning an education in biotechnology from RIT sparked my interest in learning more about the genetic basis of disease. On a personal level, I抳 e gained quite a bit from my cooperative experience. Confident in my ability to adapt to new environments and learn techniques that I have never encountered, I have also learned a great deal about the nature of private-sector research and the lifestyle of the researchers. Familiar with the amount of dedication and hard work essential to project advancement, I am more focused on my goal of becoming an excellent researcher than I ever have been. No stranger to the frustration and disappointment inherent in research work, I am spurred on by the thrill of discovery. I find a great deal of enjoyment working in research and plan to stay at Astra through the summer of 2005.I feel that my course work as a Biotechnology major at RIT and my research experience at Astra have adequately prepared me for graduate study. Eager to continue my education and improve upon my

weaknesses, I am particularly interested in becoming more familiar with emerging technologies relating to genetics and in gaining more exposure in working with eukaryotic genomes. Most importantly, I want to gain the knowledge and skills necessary to design and perform research according to my interests in genetics and disease. With these broad interests and specific goals, I am very attracted to the Graduate Program in Genetics at Duke University. With great flexibility in choosing a degree, the program would suit me well. At Duke, I would welcome the opportunity to explore my interests in Cell Biology, Neurobiology, Pharmacology, and Cancer Biology. I am also interested in learning more about possible involvement with the Duke Center for Human Genetics. My motivation to succeed is reflected in both my academic standing and my dedication to research. I feel that these attributes, combined with a profound interest in the Biological

Sciences, will ensure me success as a graduate student at Duke University.

Why should I select biology as my career? Born in a medical family for generations, I was very interested in all over around, especially in living organisms and I came into a habit of watching, touching, feeling and

protecting them who become my favorite friends later. Life science is much more intriguing and fascinating to me when I made endeavor to represent Shandong Province 400,000 high-school students to go to Beijing to take part in the National Biological Olympic Context, a Pick-Out for the International Biological Olympic Context. As a result, I selected Biotechnology, the foremost interesting field in Biology, as my college major when I won acceptance into Department of Biotechnology in the College of Life

Science at Peking University, the best university of China.

Upon acceptance to Peking University in 1996, I was awarded consecutive scholarships for all 4 years in college. During my college years, with the systematic education, I obtained a solid academic background

in chemistry and basic biology science especially in experiment skills as a preparation for future advanced research, which is also, the basis for me to do my current research project at the lab. Due to my outstanding experiment skills, in the third year of my undergraduate study, I was recommended to the State Key Lab of Microbial Resource, Institute of Microbiology, Chinese Academy of Science, which only 2% the students can get in, to do research on the structure and character of a new family of DNA-Binding Protein from the Hyperthermophilic Archaeon Sulfolobus Shibatae(extreme environment). And independently I have been dedicating in the interaction between DNA and DNA-Binding Protein by Gel Retardation Assay, Nick Closure Assay and EMSA. This project provided

me with not only considerable experiment experience including bacterial culture, expression of recombinant, and western blotting etc, but also how to plan an experiment, by consulting papers, seeking

optimal conditions and selecting appropriate methods. Now my partners and I have made a great achievement of this project and we have a paper, “A Highly Conserved DNA-Binding Protein from the

Hyperthermophilic Archaeon Sulfolobus Shibatae Affects DNA Supercoiling in a

Temperature-Dependent Fashion”, which will be published in the NUCLEIC ACIDS RESEARCH(4.488)

in the nearer future.

Furthermore, I have once, as a co-operator, helped other group investigate Archaeon ssh7 DNA binding protein and DNA interaction. With the experiment process, I am proficient in PCR and RT-PCR, southern and northern blotting, cell in-situ blotting, CsCl density gradient centrifuge, expression of recombinant

protein, FPLC, protein purification and protein/enzyme activity assay, etc

In addition to academic studying and lab training, I benefit much from my college life. I joined the Green Life Association, one of the best student organizations in Peking University, and had been the minister of propagation for one year. The job has deepened my love for lives and improved my responsibility both for

group and for the well-being of our earth. Moreover, all the many-sided pleasant experiences and excellent records are a great treasure to me, giving me high confidence and independence. Why to go abroad to pursue Ph.D.? I’m always thinking that the research of life sciences is not only interdisciplinary but also international. Besides the advanced technology, I hope to fuse the analytic way of the east and west. So I’d like to go abroad for advanced studies after my graduation in July of 2000. And besides, it is to make more research achievement in the life science and at the same time, fulfill my keen dream of standing on the foremost frontline in biology science. Why to choose your University as

my graduate school? It is that she is one of the most highly regarded universities in the world and is an ideal place for ambitious youth. Why to select your program? With my experiment skills and broad academic background, I am more and more certain that I am well prepared for the further exploration into your respected program, which is very promising in the modern biology. I wish to study Biology because of my enthusiasm for the environment and my fascination with how living organisms work, particularly animals. I enjoy studying creatures at a cellular level as well as on a large scale, such as their ecosystem. It is this variation of perspective that makes Biology an appealing subject

to further my knowledge at University.

I recently attended lectures on biodiversity and conservation at the University Museum of Natural History in Oxford. This aided my understanding of different topics, including why our rainforests are so important

and the role of specific invertebrates in our global environment.

I currently own a shared website on which my AS Biology notes can be viewed. This was set up in order to improve my skills in computer and data handling, as well as share information with other students who are studying Biology at A-level. I am hoping it will also make the subject more interactive and exciting for

people who visit the site.

I keep up to date with new developments and ideas by reading around my school subjects in books and in journals such as 'New Scientist'. Leisure activities, including visits to Cotswold Wildlife Park, the Natural History Museum and the Body world Exhibition, have proved valuable in expanding my biological

knowledge, as has watching wildlife documentaries on television.

I have a part time job at Summertown Library, which has been an important experience for me. It has not only made me more independent, but has taught me the importance of decision-making, how to work as a team and how to handle responsibility. I take pleasure in reading novels, spending time with friends,

drawing, painting and walking in the countryside.
