TNNT3突变转基因小鼠筛选及肌钙蛋白表达的研究

【摘要】Objective: To study the troponinTNNT3( R69H )gene expression and tissue structure change of fast skeletal muscle in TNNT3 mutation transgenic mouse. Methods: The heterozygotic TNNT3 mutation transgenic mouse was mated with same brood heterozygotic mouse. The positive progeny was detected by wild C57BL/6J mouse. Fast skeletal muscle of wild mouse, heterozygotic TNNT3 mutation transgenic mouse and homozygous TNNT3 mutation transgenic mouse were tested by to RT - PCR, WESTERN and electron microscope section. Results: The electropho-resis band arrange of RT - PCR, WESTERN from bright to dark were homozygous mutation transgenic mouse, heterozygous mutation transgenic mouse, wild C57BL/6J mouse. The electron microscope section of the fast skeletal muscle of TNNT3 mutation transgenic mouse displayed band derangement, edema, vacuole. Conclusion: The quantity of mRNA and protein of TNNT3 gene in homozygous mutation transgenic mouse is higher than heterozygous mutation transgenic mouse, and heterozygous mutation transgenic mouse is higher than wild C57BL/6J mouse.
